It is possible that the plug is in the vent itself. It might be worth a visit to your roof to see if there are leaves and debris, or even some animal nest, plugging the vent up top.

You can pour water down the vent to see if it backs up. If this is the case, you might be able to use a plumbing auger to remove the clog. If none of these things work, it might be time to call a plumber.
